If you purchase blank signs from Hobby Lobby, can you etch and score on them with the frame still on the sign. In other words, will the raised frame get in the way and cause damage to the head?

Yes or ruin the cut,
Anything over a half-inch above the crumb tray will get hit by the carriage fan shroud. If your surface is below it will not cut properly
It all depends on the purchased signs you are talking about. I don’t shop at Hobby Lobby so I have no idea what the product looks like. Perhaps share a photo of the item so users can give you tips on how best to engrave it.
In general, the surface to be engraved must be within the 1/2" focus range of the Glowforge. If there is a lip or frame surrounding the area you wish to engrave, it must not exceed 1/2". For materials thicker than 1/2", you will need to remove the honeycomb tray and then elevate your item so that the surface to be engraved is within 1.5" and 2" from the floor of the Glowforge.
